O prémio instituído pela Fundação BIAL tem um valor de 300 mil euros e visa reconhecer o que de mais notável e relevante tem sido descoberto na área biomédica.

O estudo ‘Cyclooxygenase-Dependent Tumor Growth through Evasion of Immunity’ distinguido com o BIAL Award in Biomedicine, publicado na revista Cell, e que envolveu um total de doze investigadores de várias nacionalidades, insere-se na área da imunologia tumoral, que serve de base para a imunoterapia do cancro, vista atualmente como potencial tratamento de primeira linha para vários tumores e decisiva no tratamento do cancro. Esta estratégia baseia-se na potenciação do sistema imunitário individual de cada paciente para combater as células cancerígenas e transformou, na última década, o tratamento de vários cancros metastáticos resistentes às terapêuticas convencionais, nomeadamente à quimioterapia.

Tendo como ponto de partida o facto de apenas uma minoria dos doentes obter respostas completas e duradouras à imunoterapia, o grupo liderado por Caetano Reis e Sousa analisou as causas de insucesso terapêutico e identificou uma possível explicação para o mecanismo de “fuga” tumoral ao sistema imunitário e consequente resistência à imunoterapia: a enzima ciclooxigenase e um dos seus produtos principais, o lípido inflamatório prostaglandina E2. Estas moléculas são essenciais para que as células cancerígenas consigam escapar ao reconhecimento e eliminação pelo sistema imunitário. Quando manipularam geneticamente a capacidade das células tumorais produzirem estas moléculas, os autores observaram um ataque imunitário maciço aos tumores, que perderam volume até desaparecerem.

A equipa distinguida estudou ainda a capacidade dos inibidores de ciclooxigenase, utilizados nas doenças inflamatórias, caso da aspirina, de aumentar a eficácia da imunoterapia no tratamento de vários cancros, como o melanoma ou os carcinomas do pulmão e do rim. Os resultados mostram um aumento notável de eficácia da combinação da aspirina com a imunoterapia, em relação aos tratamentos individuais, resultando na erradicação dos tumores nos modelos pré-clínicos em estudo.

A vice-presidente do júri do BIAL Award in Biomedicine, Maria do Carmo Fonseca, salienta que “este trabalho pioneiro inspirou novos ensaios clínicos, atualmente em curso à escala mundial, em que a combinação de fármacos anti-inflamatórios como a aspirina com a imunoterapia está a ser testada em vários tipos de cancros, tais como os da mama e do cólon”.

 

Caetano Reis e Sousa confessa que “foi com grande alegria que recebi um telefonema do júri a anunciar que um estudo do meu laboratório tinha sido o vencedor do BIAL Award in Biomedicine 2019. Trata-se de uma investigação pré-clínica sobre um dos mecanismos utilizados pelos cancros para evitar a deteção do sistema imunitário. O estudo foi realizado por uma equipa de talentosos investigadores do nosso laboratório e colaboradores externos e estamos todos muito entusiasmados e honrados por receber este prestigiante prémio. Para mim, como cidadão português, receber o BIAL Award in Biomedicine tem um significado especial. Em nome de todos os que participaram no estudo, agradeço ao júri por escolher o nosso trabalho e à Fundação BIAL que patrocina o prémio.”

Caetano Reis e Sousa nasceu em Lisboa, em 1968, e foi viver para o Reino Unido na adolescência. Licenciou-se em biologia no Imperial College de Londres e fez o doutoramento em imunologia na Universidade de Oxford. No ano passado, Caetano Reis e Sousa foi eleito membro da Royal Society, tornando-se o primeiro português em 200 anos a entrar como fellow para a academia de ciências britânica, a mais antiga do mundo. Em 2009, Caetano Reis e Sousa foi distinguido com a Ordem portuguesa de Sant’Iago da Espada e em 2017 recebeu o Prémio Louis-Jeantet de Medicina.
